Artem Dzyuba's sensational chip cleaned up a messy scramble in spectacular fashion to set Zenit on the path to a title-winning performance.
Sardar Azmoun got in on the act towards the end of the first half, glancing home a fine cross from Sebastian Driussi. In added time before the break, he converted a penalty to put the champions 3-0 up.
Within seconds of the restart, he completed his hattrick with a fine volley from Malcom's deep cross. Five minutes later Dzyuba tapped in a rebound from the crossbar to make it five.
The slenderest of hopes was granted to Lokomotiv when François Kamano chased down Yaroslav Rakitsky dribbling across the six yard box, and slotted home after winning possession. The hope was extinguished though 10 minutes later when Malcom audaciously chipped Guilherme from well outside the box.
Zenit are crowned champions after going nine points clear of Lokomotiv in second.
