RPL champions Zenit were ruthless in picking off Lokomotiv as goals from Daler Kuzyaev, Sardar Azmoun and Aleksandr Erokhin all came from sloppy errors.
Daler Kuzyaev was alert to intercept Grzegorz Krychowiak's weak pass intended for Dmitry Barinov, before striding towards goal and picking his spot clinically from the edge of box.
Rifat Zhemaletdinov and Sardar Azmoun both had good chances in the opening exchanges of the second half. The Lokomotiv forward steered a low shot just wide moments after last season's RPL player of the season had stung Guilherme's gloves with a powerful shot.
Azmoun doubled the lead after 56 minutes after a sumptuous Wendel through ball evaded Pablo's desperate attempt to intercept. The Iranian calmly strolled around the stranded Guilherme and walked the ball into the net.
Lokomotiv's veteran keeper was again in the spotlight with seven minutes remaining as his howler gifted a third Zenit goal. Guilherme miscontrolled a simple Pablo back-pass a few yards out, leaving Artem Dzyuba free to set up Erokhin to smash it home from close range.
The scoreline could have been more emphatic, but Magomed Ozdoev's late effort smacked the bar and went over after Malcom's mazy dribble set up his chance.
Both sides begin their RPL campaign on Saturday 24 July, when Zenit travel to take on Khimki and Lokomotiv host Arsenal Tula.
