The draw for the first knockout round of the 2021/22 Russian Cup took place in the Match TV studios.

Nine Russian Premier Liga teams will take part in the last 16: Zenit, Spartak, Lokomotiv, Rubin, Sochi, Dynamo, CSKA, Nizhny Novgorod and Arsenal. Six more participants represent the second-tier FNL: Kuban, KAMAZ, Alania, Rotor, Baltika, Yenisei. The remaining spot in the draw is taken by the sole third-tier entrant remaining, FNL-2 side Chaika.

Preliminary match dates are 1-3 March 2022.

The current holders of the trophy are Lokomotiv Moscow.
